Is mirror lock-up one of the keys to getting a better macro shot? I am under the impression that a tripod and a shutter release cable really helps. Also...there are some focus rails for this type shot..which are probably a good investment if you really enjoy macro shots.

They help only if you are using available light. If you use flash for your exposure it doesn't make any difference
Flash does two things.
One, flash will freeze your image the moment your flash pops, independent of your shutter speed. The flash is much faster then your camera will ever be.
Two, flash will allow you to set your aperture to your highest value enabling you a greater depth of field. (example: f/42). Try that with available light settings and you'll be exposing that one shot all day long (so to speak).
If you are not using flash or using flash with ambient, then yes, anything that will steady your camera will definitely help.
But to get those crystal clear shots, use flash.
A focus rail is a good idea if you have one. Depending on how close you get to the subject, your depth of field can be very small. A 16th of an inch can make a big difference in nailing that focus point and its a lot easier then trying to move you tripod back and forth. It's something you don't absolutely need, but nice to have.
The problem is, like all the other equipment I have, it's something else I would have to carry around and usually wouldn't have it with me when I wanted to use it unless it was planned in advance.
But if you want to be a macro shooter, you should have a cable release and a focus rail. If it's just an occasional macro shot now and then it may not be worth it.

I'm not going to get into which lens's are the best. Thats a matter of opinion and probably open up a can of worms.
The best place to get a lot of opinions on this will be dpreview. Ask there and you'll get your answer or do a search on macro in the dpreview lens forum. That question has been asked many times before and the answer varies.

I use my general purpose lens (Tamron 28-300). I can focus down to about a foot anywhere in the 28-300 range. I took the below (Link) pictures with it and they came out fine. Not as sharp as I would have liked it but never the less, acceptable.

Most likely the sharpest lens you can get will be a primary lens (No Zoom). I shy away from those because they are limited use lens and I tend not to have them with me when out of the house.
I should have the Tamron with me tonight if you want to look at it. Its an excellent general purpose lens for what it does. Duane Whitaker has one he's selling for about $150. It's on Ebay right not so that means he will probably take around $135 (Less ebay/paypal fees) Mine cost me $700 but its the image stabilized version. I also used the tamron to shoot the below (link) pictures.

And a final note.
When ever I run across a website that has pictures that interest me such as this, I save them to my computer for later study. If I know Im going to be shooting a particular subject, it helps to prep before hand and study the saved images for the shoot. I then plan and equip myself with the necessary equipment. Basically, I know what I want to accomplish before I leave. I give myself a goal.
It’s simple enough to save them to my hard drive but in some cases its easier to snatch them from the screen. Some sites don’t allow saving and I find this method easier then saving because it ends up being faster in the long run. Anyway, I thought I would give you a little bonus for using the forum and turn you on to a little program I use to do just that. Its called HoverSnap (PC) and its free.
